摘要:
观察有氧运动对APOE-/-小鼠皮下脂肪组织(SAT)、内脏脂肪组织(VAT)和心外膜脂肪组织(EAT)脂代谢、氧化应激和炎性状态的影响;探索不同脂肪组织中与脂代谢、氧化应激和炎性相关的蛋白对有氧运动的适应性特征。实验步骤:把16只雄性APOE-/-小鼠随机分为安静组和运动组。12wk有氧运动干预后,检测SAT、VAT和EAT的FABP4、perilipin、GSH-Px、Cu/Zn-SOD、Mn-SOD、adiponectin、VEGFα、TNF-α和IL-6蛋白表达量。实验结果:(1)有氧运动明显增加VAT、EAT的FABP4的表达(P<0.01),SAT的FABP4无明显变化(P>0.05);有氧运动导致VAT的perilipin明显降低(P<0.01),EAT的perilipin有降低的趋势(P>0.05),SAT的perilipin无明显变化(P>0.05);(2)有氧运动明显增加VAT、EAT的GSH-Px(P<0.01)和Mn-SOD的表达(P<0.05),Cu/Zn-SOD无明显变化(P>0.05);有氧运动对SAT的GSH-Px、Cu/Zn-SOD和Mn-SOD无明显影响(P>0.05);(3)有氧运动导致VAT的adiponectin明显增加(P<0.01),EAT的adiponectin有增加的趋势(P>0.05),SAT的adiponectin有降低的趋势(P>0.05);(4)有氧运动导致SAT、VAT和EAT的IL-6明显降低(P<0.01),VAT、EAT的TNF-α明显降低(P<0.01),SAT的TNF-α无明显变化(P>0.05);(5)有氧运动导致SAT的VEGFα明显增加(P<0.01),而VAT、EAT无明显变化(P>0.05)。结论:(1)有氧运动有效改善了APOE-/-小鼠VAT、EAT的脂代谢、氧化应激和炎性状态,对SAT影响不明显(除IL-6降低,VEGFα升高);(2)VAT、EAT中脂代谢、氧化应激和炎性相关蛋白对有氧运动表现出较为相似的生物学适应;(3)而SAT表现特征与VAT、EAT不同(除IL-6,Cu/Zn-SOD)。
Abstract:
Objective:To observe the effect of aerobic exercise on lipid metabolism,oxidative stress and inflammatory state of subcutaneous adipose tissue (SAT),visceral adipose tissue (VAT) and epicardial adipose tissue (EAT) in APOE-/- rats,at the same time,the adaptive changes of lipid metabolism,oxidative stress and inflammatory proteins in different adipose tissues were investi gated.Method:16 male APOE-/- rats were randomly divided into two groups:quiet group and exercise group.the expression of FABP4,perilipin,GSH-Px,Cu/Zn-SOD,Mn-SOD,adiponectin,VEGF-α, TNF-αand IL-6 in SAT,VAT and EAT were detected after 12wk aerobic exercise.Results:1)The expression of FABP4 in VAT and EAT was significantly increased (P<0.01),there was no significant change in SAT (P>0.05) after aerobic exercise;The expression of perilipin was significantly lower in VAT(P<0.01),the perilipin in EAT has a decreasing trend(P>0.05),the perilipin in SAT had no obviously change (P>0.05) after aerobic exercise;2)The expression of GSH-Px(P<0.01)and Mn-SOD (P<0.05) was significantly increased,and there was no significant change in Cu/Zn-SOD (P>0.05) in VAT and EAT after aerobic exercise;There were no significant effect on GSH-Px,Cu/ZnSOD and Mn-SOD in SAT (P>0.05) after aerobic exercise;3)Aerobic exercise resulted in a significant increase in adiponectin in VAT (P<0.01), the adiponectin of EAT has an increasing trend (P>0.05),and the adiponectin in SAT has a decreasing trend (P>0.05);There were a significant reduction of the expression of IL-6 (P<0.01) in SAT,VAT and EAT,a significant decrease of the expression of TNF-α in VAT and EAT(P<0.01),and no significant change in SAT (P>0.05) after aerobic exercise;Aerobic exercise resulted in a marked increase in VEGF-α (P<0.01) in SAT while no significant changes in VAT and EAT were observed (P>0.05).Conclusions:1)Aerobic exercise effectively improved the lipid metabolism,oxidative stress and inflammatory state of VAT and EAT but had no obviously effect on SAT(except IL-6 which went down,VEGF-a which went up)in APOE-/- rat;2)The research showed relatively similar biological adaptation about the related proteins of lipid metabolism,oxidative stress and inflammatory response to aerobic exercise in VAT and EAT while SAT showed biological adaptation was different from VAT and EAT (except IL-6 and Cu/Zn-SOD ).
