摘要:
克隆了大黄鱼(Larimichthys crocea)a20基因(Lca20)的编码区序列,采用实时荧光定量PCR(qRT-PCR)检测了Lca20在大黄鱼不同组织中的表达水平,以及免疫刺激后的表达变化。结果显示:Lca20包含2475 bp的开放阅读框,编码824个氨基酸;预测其蛋白质包含1个OTU结构域和6个ZnF结构域;进化分析表明,LcA20与来自其他硬骨鱼的A20亲缘关系较近;qRT-PCR显示,Lca20广泛分布于大黄鱼多个组织,在脾脏组织中表达量最高,在肌肉组织中表达量较低;LPS和Poly I∶C刺激可以诱导大黄鱼LCK细胞中的Lca20表达量显著增加,最大值约分别为对照组的6.20倍(12 h)与7.73倍(24 h)(P<0.01);杀香鱼假单胞菌攻毒也可以诱导大黄鱼脾脏、头肾等主要免疫组织中的Lca20表达量显著增加,其中,在攻毒后24 h,头肾中的Lca20表达量增加了7.87倍(P<0.01),提示Lca20可能在大黄鱼抵抗不同病原感染的过程中发挥重要作用。
Abstract:
In this study,the sequence of the coding region of the a20 gene(Lca20) of Larimichthys crocea were cloned,and the expression level of Lca20 in different tissues of L.crocea and the changes of expression after immune stimulation by quantitative real-time PCR(qRT-PCR)were detected.The results showed that Lca20 contains an open reading frame of 2475 bp,encoding 824 amino acids.The predicted Lca20 protein contains one OTU domain and six ZnF domains.Phylogenetic analysis showed that LcA20 was clustered to the teleosts,A20 subgroup.The qRT-PCR analysis indicated that Lca20 was widely distributed in the examined tissues of L.crocea,with the highest expression in spleen tissue and the lowest expression in muscle tissue.LPS and Poly I:C stimulation induced a significant increase in the expression of Lca20 in LCK cells of L.crocea,with the highest values of 6.20 times(12 h) and 7.73 times(24 h) higher than those of the control group,respectively(P<0.01).Intraperitoneal injection of Pseudomonas plecoglossicida also induced a significant increase in the expression of Lca20 in most immune tissues of L.crocea,reaching the peak value of 7.87 times higher than that of the control in headkidney tissue at 12 h after infection.This results suggested that a20 might play an important role in L.crocea immune response.
