克隆了一种大黄鱼baff-样(Lcbaff-like)基因的cDNA序列,采用实时定量PCR(qPCR)检测了该基因在大黄鱼不同组织中的表达情况,以及免疫刺激后的表达变化。序列分析显示,Lcbaff-like cDNA序列包含长度为669 bp的开放阅读框(ORF),编码222个氨基酸,预测其蛋白质包含1个TNF结构域。进化分析表明,LcBaff-like与来自其他硬骨鱼的Baff-like聚为一支,并与鮸鱼(Miichthys miiuy) Baff-like同源性最高。qPCR显示,Lcbaff-like在大黄鱼血细胞中表达量最高,在肌肉组织中表达量最低。受到LPS与Poly (I:C) 刺激后,大黄鱼LCK细胞系中该基因表达量极显著增加,最大值分别为对照组的6.20倍(刺激后6 h)与7.16倍(刺激后12 h)(P<0.01)。采用变形假单胞菌人工感染后,大黄鱼鳃、皮肤、脾脏及头肾中的Lcbaff-like均被显著诱导,最大值分别为对照组的3.89、2.44、7.09与5.41倍(P<0.01)。研究结果提示Lcbaff-like可能参与大黄鱼抵御细菌与病毒感染的免疫反应。
Abstract:
The cDNA sequence of a large yellow croaker baff-like (Lcbaff-like) gene was cloned.The expression levels of it in various tissues of the large yellow croaker,as well as its dynamic changes in vivo and in vitro following immune challenge,were analyzed by real-time quantitative PCR (qPCR).The results showed that the Lcbaff-like cDNA sequence contained a 669 bp open reading frame,encoding 222 amino acids.The predicted LcBaff-like protein consisted of a conservative TNF domain.Phylogenetic tree analysis indicated that LcBaff-like was clustered to the same subgroup with the BAFF from other teleost,sharing the closest relationship with the Baff-like from Miichthys miiuy.qPCR analysis revealed that Lcbaff-like existed in all examined tissues of the large yellow croaker,with the most predominant expression in blood cells,followed by the gills and spleen,and the weakest in muscle tissue.Upon stimulation with LPS and Poly (I:C),the expression levels of Lcbaff-likein LCK cells increased significantly,reaching the peak value of 6.20 and 7.12 times as much as that of the control at 6 h and 12 h,respectively (P<0.01).After infection with Pseudomonas plecoglossicida,Lcbaff-liketranscripts were induced in most immune tissues of large yellow croaker,with the highest level of 7.09 times as much as that of the control appeared in the spleen at 24 h post-infection (P<0.01).These findings suggest that Lcbaff-likemight play a role in the immune response of the large yellow croaker against bacterial and viral infections.
