摘要:
建立高效液相色谱法(high performance liquid chromatography,HPLC)快速测定藻油中二十二碳六烯酸乙酯(docosahexaenoic acid ethyl ester,DHA-EE)含量的方法。将藻油经乙酯化处理后,采用优化后DHA-EE分析参数:色谱柱ZORBAX SB-C8(4.6 mm×250 mm),流动相V(甲醇)∶V(水)=90∶10,柱温为30 ℃,流速为1.0 mL/min,紫外检测波长为204 nm,进样量为10 μL。测定结果显示,DHA-EE在0.008 1~1.92 μg 范围内呈良好线性关系,线性回归方程为:y=39.91 x+765.11(R2=0.999 2),平均回收率为102.42%(n=5,RSD=2.25%),精密度(n=10,RSD=0.32%)较好,DHA-EE最低定量限达8.05 ng。表明,该方法可用于藻油及其制品中DHA-EE的检测分析。
Abstract:
High performance liquid chromatography(HPLC)method was developed to quantify DHA-EE in marine microalgae oil quickly.The optimized quality parameters for DHA-EE analysis were as follows:chromatographic column,ZORBAX SB-C8(4.6 mm×250 mm);mobile phase,methanol versus water ( 90∶10);the column temperature 30 ℃;the flow velocity 1.0 mL/min;UV wavelength 204 nm;the injection volume 10 μL.It presented a good linear relationship within range of 0.008 1-1.92 μg for DHA-EE,and the linear regression equation was y=39.91 x+765.11(R2=0.999 2). The average recovery rate was 102.42% (n=5,RSD=2.25%) with good precision(n=10,RSD=0.32%),and the minimum quantify limit reached 8.05 ng.This optimized HPLC method could be used in the determination and analysis of algal oil and DHA-EE
