摘要:
为探究深海细菌Flammeovirga pacifica降解龙须菜所产生的寡糖抗食物过敏功效,以龙须菜为原料,通过发酵、超滤、Superdex Peptide HR 10/300柱层析等方法获得高纯度寡糖.离子色谱结果显示:分离纯化获得的寡糖主要为四糖和六糖,其质量分数分别为48.47%和25.69%.小鼠过敏模型实验结果表明,腹腔注射该寡糖(100 μg/只)能显著降低过敏小鼠血清特异性IgE和IgG1水平,提高特异性IgG2a水平.通过灌胃(10 mg/只)能显著降低小鼠粪便中组胺的水平,且能有效缓解空肠组织过敏病理现象.小鼠脾淋巴细胞实验结果显示,该寡糖能在转录和转译水平上显著降低Th2细胞因子(IL-4、IL-5、IL-13)含量,促进Th1细胞因子IFN-γ分泌.说明该寡糖能有效调节辅助性T细胞的免疫功能向Th1细胞转换,从而抑制Th2细胞主导的食物过敏反应.
Abstract:
In order to investigate the anti-food allergic effect of Gracilaria lemaneiformis oligosaccharides which were degraded by the deep sea bacteria Flammeovirga pacifica,the oligosaccharides were obtained from Gracilaria lemaneiformis through fermentation,ultrafiltration,and purified by Superdex peptide HR 10/300 chromatography column.The oligosaccharides obtained are mainly tetrasccharide and hexaose,their contents were 48.47% and 25.69% respectively as detected by ion chromatography.The results of the allergic mouse model indicated that intraperitoneal injection of Gracilaria lemaneiformis oligosaccharides (100 μg/mouse) significantly suppressed serum specific IgE,IgG1 level,and promoted specific IgG2a level in mice.Gavage of Gracilaria lemaneiformis oligosaccharides(10 mg/mouse) decreased fecal histamine release and repaired the pathology in the jejunum.The results of the mouse splenic lymphocytes experiment showed that Th2 cytokine (IL-4,IL-5 and IL-13) contents were reduced,while the Th1 cytokine (IFN-γ) content was up-regulated by Gracilaria lemaneiformis oligosaccharides in the transcription and translation levels.These results suggested that Gracilaria lemaneiformis oligosaccharides could cause the immune function of helper T cells to convert to the Th1,so as to inhibit the food allergies dominated by Th2 cells.
