以钝顶螺旋藻(Spirulina platensis)干粉为实验原料,经过超滤、两步盐析、DEAE离子交换色谱柱层析和SuperdexTM200凝胶过滤柱层析,逐步纯化超氧化物歧化酶(superoxide dismutase,SOD)。结果表明,在90 L粗藻液的中试放大体系中,所得酶液的平均比活力为4 131 U/mg,纯化倍数为238.96,回收率为8.3%。进一步利用十二烷基硫酸钠-聚丙烯酰胺凝胶电泳(sodium dodecyl sulfate polyacrylamide gel electrophoresis,SDS-PAGE)实验,确定所得的SOD蛋白质分子质量约为18.4 ku。此外,3次重复实验获得的SOD酶活力均大于4 000 U/mg。
Abstract:
In this study,superoxide dismutase (SOD) was isolated from Spirulina platensis,and then sequentially purified through ultrafiltration,two-step salting-out method,DEAE ion exchange chromatography and SuperdexTM200 gel filtration chromatography.The results showed that the average specific of the enzyme was 4 131 U/mg,the purification multiple was 238.96 and the extraction recovery of this protein was 8.3% in a pilot scale system of 90 L crude algal extract.The result of SDS-PAGE analysis demonstrated that molecular mass of SOD was 18.4 ku.Repeat experiments showed similar results.In addition,the SOD activity of three repeated experiments was all greater than 4 000 U/mg.
