以钝顶螺旋藻(Spirulina platensis)藻粉为原料,采用热碱浸提法提取螺旋藻中多糖类化合物。通过单因素实验及正交试验对螺旋藻多糖的提取工艺进行优化,用等电点沉淀与离子交换色谱分离技术联用工艺进行脱蛋白,并与Sevag法脱蛋白作对比。同时,对比探究了不同浓度下螺旋藻多糖的DPPH自由基的清除能力及其对正常细胞LO2和肿瘤细胞HepG2增殖的抑制作用。结果表明,影响最为显著的因素为提取时间,当料液比(m∶V)为1∶20、浸提温度为80 ℃、浸提时间为3 h、碱液质量分数为1.25%时,多糖提取率为8.78%;等电点沉淀与离子交换树脂分离技术联用工艺法的多糖得率(7.55%)及蛋白质脱除率(89.54%)均高于Sevag法的多糖得率(6.33%)及蛋白质脱除率(48.40%);螺旋藻多糖对DPPH自由基具有较好的清除作用;500~2 000 mg/L质量浓度范围内的螺旋藻多糖对LO2细胞无毒,对HepG2细胞有显著的抑制作用。

The single-factor test and orthogonal array design were used to optimize polysaccharide extraction from Spirulina platensis by hot alkali method.The efficiencies of various deproteinization methods including Sevag and isoelectric point precipitation combined with ion exchange chromatography were compared.The DPPH free radical scavenging ability of Spirulina polysaccharides was measured in vitro.The inhibitory effect of Spirulina polysaccharide on normal cell LO2 and tumor cell HepG2 proliferation was tested.The results showed that extraction time was the main factor affecting the polysaccharide yield,and the best extraction conditions were as follows:material -liquid ratio of 1∶20,extraction temperature of 80 ℃,extraction time of 3 h,alkali concentration of 1.25%.Under the above conditions,the yield of polysaccharide was 8.78%.Isoelectric point precipitation combined with ion exchange chromatography method was more efficient than the Sevag method.Spirulina polysaccharide had a good scavenging effect on DPPH free radicals and it had no effect on the cell proliferation ability of LO2.Besides,it significantly inhibited the proliferation of liver cancer cells HepG2 at 500 -2 000 mg/L.
