摘要:
为探究中草药对原代培养花鲈肝细胞氧化损伤的保护作用,选取10种复方(记为M1、M2、M3、M4、M5、M6、M7、M8、M9、M10)和10种单方中草药,煎制成终质量浓度为5 mg/mL的水提物,用这20种水提物处理花鲈原代培养肝细胞4 h,再换用200 μmol/L H2O2处理2 h进行氧化损伤,最后测定培养液中胞内酶活性(AST、ALT和LDH)及肝细胞抗氧化指标(T-AOC、SOD和MDA)。结果显示:肝细胞受损后,其培养液中AST、ALT和LDH的活性显著升高(P<0.05),肝细胞中T-AOC和SOD活性显著降低(P<0.05),MDA含量显著上升(P<0.05);先经中草药处理再用H2O2氧化损伤后,各中草药处理组AST、ALT和LDH活性降低,其中姜黄、五味子单方和M9复方的AST、ALT和LDH活性较其他中草药组显著降低(P<0.05);姜黄单方和M9复方的T-AOC和SOD活性较其他组明显升高(P<0.05),MDA含量显著降低(P<0.05)。由此得出:大黄、丹参、银杏、姜黄、甘草、赤芍、猪苓、五味子单方,M1、M4、M6、M7、M9复方均能缓解由H2O2诱导的原代培养花鲈肝细胞损伤,其中姜黄单方和M9复方的保护作用最强。
Abstract:
To explore the protective effects of Chinese herbal medicine on oxidative damage of Lateolabrax maculatus hepatocytes in primary culture,ten kinds of single traditional chinese medicine and ten kinds of compound Chinese medicine (M1,M2,M3,M4,M5,M6,M7,M8,M9 and M10) were selected and made into water extract whose final concentration was 5 mg/mL.The primary hepatocytes were treated with 20 kinds of water extract for 4 h,and then treated with 200 μmol/LH2O2 for 2 h to cause oxidative damage.The results of this study showed that H2O2 treated hepatocytes exhibited significant increases of AST,ALT and LDH in culture medium and the content of MDA in hepatocytes (P<0.05),while the activities of T-AOC and SOD were significantly decreased(P<0.05).Adding Chinese herbal medicine for protection before oxidation damage with H2O2,the activity of AST,ALT and LDH were decreased in each herbal treatment group,while AST,ALT and LDH in Curcumae longae,Schisandrae chinensis and M9 group were significantly lower than others Chinese medicine groups (P<0.05).T-AOC and SOD in Curcumae longae and M9 groups were higher than other groups (P<0.05),and MDA content was significantly reduced (P<0.05).To sum up,among the 10 single formulae and 10 groups of compound Chinese herbal medicines in this study,Chinese rhubarb,Salvia miltiorrhiza,Ginkgo biloba,Curcumae longae,Glycyrrhiza uralensis,Paeonia,Polyporus,Schisandra chinensis,M1,M4,M6,M7 and M9 could all alleviate the hepatocyte injury induced by H2O2 in primary culture of L.maculatus,while Curcumae longae and M9 groups have the strongest protective effect.
