]由变形假单胞菌(Pseudomonas plecoglossicida)引起的大黄鱼(Larimichthys crocea)内脏白点病的死亡率可达50%~80%。对抗病相关基因trim38进行了分子及表达特征等分析。结果显示:该基因开放阅读框(ORF)1623 bp,编码540个氨基酸,预测蛋白分子量为61.58 ku,等电点6.99。qRT-PCR的结果表明,在所有检测的组织中都发现了trim38的转录产物,但在肾脏中的相对表达量最高。变形假单胞菌攻毒刺激后,大黄鱼脾脏中该基因持续表达,在120 h达到峰值;肝脏中3 h显著表达,之后相对表达量降低;肾脏中该基因相对表达量变化不大,在96 h左右达到最大值。上述结果表明trim38基因参与了大黄鱼应对变形假单胞菌感染的抗病免疫过程。
Abstract:
Visceral white spot disease caused by Pseudomonas plecoglossicidais one of the main diseases of large yellow croaker cultured.The infection rate reaches more than 50%.In this study,a candidate diseaseresistancerelated gene trim38was screened from the above-located major regions,and its molecular and expression characteristics were analyzed.The results showed that the gene open reading frame (ORF) was 1623 bp,encoding 540 amino acids,the predicted protein molecular weight was 61.58 ku,and the isoelectric point was 6.99.The results of qRT-PCR showed that trim38 mRNA transcripts were found in all detected tissues,but the highest relative expression was in the kidney.After challenge with P.plecoglossicida,the gene was continuously expressed in the spleen of large yellow croaker and reached a peak at 120 h;it was significantly expressed in the liver at 3 h and then decreased;the relative expression of the gene in the kidney did not change much and reached the maximum at about 96 h value.These results indicated that trim38was involved in the immune process of large yellow croaker in response to P.plecoglossicida infection.
