摘要:
大黄鱼雄鱼体型和体色优于雌鱼,后期养殖成活率和市场售价也高于雌鱼,全雄苗种受到部分养殖业者欢迎,而激素诱导是实现单性化养殖的有效手段。在雌二醇(E2)中加入一定比例的曲洛斯坦(TR)作为诱导药物,将混合药物用壳聚糖制成胶条或溶解于大豆油中制成缓释液,埋植或注射到大黄鱼成熟雄鱼的腹腔进行性逆转诱导实验,观察性腺的组织学变化,检测血清中性激素浓度,检测性腺中雄性特异基因和雌激素受体基因表达量的变化。结果表明:E2-TR-大豆油混合液的诱导效果优于壳聚糖-E2-TR 药条;按每公斤鱼体重腹腔注射40 μg E2+ 4 μg TR,持续处理 80 d 后,实验组大黄鱼的精巢退化,形态变成与卵巢相似,其中观察到有大量卵原细胞,血清中11酮基睾酮浓度显著下降、雌二醇浓度显著上升(P<0.05),性腺中dmrt1基因的表达显著下调、esr2基因的表达显著上调。这说明通过诱导成熟雄性大黄鱼的性逆转,培育出生理性雌鱼是可行的。
Abstract:
Male large yellow croakers exhibit superiority over females in terms of body size and coloration,and they also tend to have higher survival rates and market prices during later stages of aquaculture.Consequently,all-male seedlings are favored by some aquaculture practitioners.Hormone induction represents an effective strategy for achieving monosex breeding in this species.To investigate induced sex reversal,a mixture of estradiol (E2) and a certain proportion of trenbolone acetate (TR) was used as the inducing agent.This mixture was formulated into chitosan strips or dissolved in soybean oil to create a sustained-release solution,which was then injected into the abdominal cavity of mature female large yellow croakers.The histological changes of the gonads were observed,and changes in serum sex hormone concentrations,expression levels of male-specific genes,and estrogen receptor genes in the gonads were detected.The results indicated that the induction effect of the E2-TR-soybean oil mixture was superior to that of chitosan-E2-TR strips.When the E2-TR-soybean oil mixture was administered at a dose of 40 μg E2+4 μg TR every 1 kg body weight via intraperitoneal injection for 80 consecutive days,the testes of the experimental group of large yellow croakers degenerated,taking on a morphology similar to ovaries with the formation of numerous oogonia.Notably,serum concentrations of 11ketotestosterone significantly decreased,while estradiol concentrations markedly increased (P<0.05).Furthermore,the expression of the dmrt1gene in the gonads was significantly downregulated,while the expression of the esr2 gene was significantly upregulated.The study indicates that inducing sex reversal in mature male large yellow croakers to develop physiological females is feasible.
