以海洋赫氏颗石藻(Emiliania huxleyi)为材料,采用超高效液相色谱串联四极杆静电场轨道阱质谱(ultra-high performance liquid chromatography coupled with quadrupole exactive mass spectrometry,UPLC-Q-Exactive-MS)技术对其细胞总脂进行定性及相对定量分析。基于UPLC-Q-Exactive-MS一级质谱扫描及二级碎片信息扫描,共鉴定出4大类232种脂质分子,包括54种甘油磷脂(26 PC,13 PG,5 PI,4 PS,2 PE,2 PA,2 CL)、142种甘油脂(21 DG,94 TG,27 MGDG)、20种脂肪酰(13 FA,5 OAHFA,2 WE)和16种鞘脂(11 Cer,5 SPH),它们分别占总脂的55%、28%、15%和2%。分析表明,赫氏颗石藻中含量最多的甘油磷脂为双磷脂酰甘油(CL),占总甘油磷脂的36.69%;甘油酯中三酰甘油(TG)的种类最丰富,占总甘油酯的32.20%;脂肪酸多为n-3多不饱脂肪酸,其中22∶6n-3(DHA)的含量占总脂肪酸含量的90.36%;鞘脂中神经酰胺(Cer)含量高达9.04 mg/g(干重),其中Cer 40∶2(isomer2)为6.28 mg/g(干重)。

The total lipid of Emiliania huxleyiwas analyzed by using ultra-high performance liquid chromatography coupled with quadrupole exactive mass spectrometry (UPLC-Q-Exactive-MS) based on orbitrap,MS/MS fragmentation pattern,and lipid species were identified.A total of 232 lipid molecules classified into four major categories were identified,including 54 glycerophospholipids (26 PC,13 PG,5 PI,4 PS,2 PE,2 PA,2 CL),142 glycerolipids (21 DG,94 TG,27 MGDG),20 fatty aclys (13 FA,5 OAHFA,2 WE) and 16 sphingolipids (11 Cer,5 SPH),which accounted for 55%,28%,15% and 2% of the total lipids,respectively.Cardiolipin (CL) is the most abundant lipid in E.huxleyi,accounting for 3669% of the total glycerophospholipids.The most abundant glycerolipid was triglyceride (TG),accounting for 32.20% of the total glycerolipids.The fatty acids were mainly n-3 polyunsaturated fatty acids,and the content of 22∶6n-3 (DHA) reached 90.36% of the total fatty acid.The sphingolipids ceramide content was 9.04 mg/g,of which Cer 40:2 (isomer2) reached 6.28 mg/g.
