摘要:
从大黄鱼的转录组数据中鉴定出两种新型c型溶菌酶,分别命名为LcLyz1与LcLyz2,对其进行序列分析、同源比对,详细研究这两种溶菌酶的蛋白质三级结构,并进行基因克隆、原核表达和蛋白质溶解性分析。结果表明,LcLyz1与LcLyz2的开放阅读框分别为429和432对碱基对,编码142和143个氨基酸序列,它们的活性中心均定位于Glu50和Asp67,并且含有信号肽和保守的HLH(helix-loop-helix)结构。通过原核表达系统,观察到这两种蛋白均以包涵体的形式在沉淀中表达。
Abstract:
In this study,two novel c-type lysozymes,LcLyz1 and LcLyz2,were identified within the large yellow croaker transcriptome.Sequence analysis,homologous alignment were performed,including an examination of their tertiary protein structures,alongside gene cloning,prokaryotic expression,and solubility assessments.The open reading frames (ORFs) of LcLyz1and LcLyz2are 429 and 432 bp,respectively,encoding 142 and 143 amino acids.Both sequences exhibit active sites located at Glu50 and Asp67,along with signal peptides and a conserved helix-loop-helix (HLH) structure.Prokaryotic expression revealed that both lysozymes were expressed in the form of inclusion bodies in the precipitate.Keywords: Larimichthys crocea;c-type lysozyme;molecular characteristics;protein expression
