利用生物信息学方法从大口黑鲈(Micropterus salmoides)基因组中鉴定出7个Dmrt基因,包括Msdmrt1、Msdmrt2a、Msdmrt2b、Msdmrt3、Msdmrt4、Msdmrt5和Msdmrt6。这些基因在染色体上分布不一,其中Msdmrt1-Msdmrt3-Msdmrt2a呈线性成簇排列。系统进化关系分析结果显示,大口黑鲈Dmrt家族与同属鲈形目的大黄鱼(Larimichthys crocea)关系较近,与其他硬骨鱼Dmrt家族的关系相对保守。结合大口黑鲈转录组数据、组织半定量PCR与荧光定量PCR分析结果显示,Msdmrt基因在性腺中呈现明显的二态性表达,其中Msdmrt1、Msdmrt3、Msdmrt4和Msdmrt6仅在精巢中表达,而Msdmrt5在脑组织和卵巢中均有表达,这种表达差异可能与各基因启动子区域所包含的转录因子结合位点不同有关。

In this study,seven Dmrt genes,including Msdmrt1,Msdmrt2a,Msdmrt2b,Msdmrt3,Msdmrt4,Msdmrt5,and Msdmrt6,were identified from the Micropterus salmoides(largemouth bass) genome using bioinformatics methods.These genes are unevenly distributed across the chromosomes,among which Msdmrt1,Msdmrt3,and Msdmrt2a are linearly clustered.Phylogenetic analysis revealed that the Msdmrt gene family of largemouth bass is closely related to that of Larimichthys crocea(large yellow croaker) from the same order,and relatively conserved compared to other teleost Dmrt gene families.Combined with transcriptome data,semi-quantitative PCR,and quantitative PCR analyses,the Msdmrt genes showed clear sexually dimorphic expression in the gonads.Msdmrt1,Msdmrt3,Msdmrt4,and Msdmrt6 were specifically expressed in the testis,while Msdmrt5 was expressed in both the brain and ovary.These differences in expression may be associated with distinct transcription factor binding sites in the promoter regions of each gene.
