从厦门市集美区近海自然海水中分离筛选出一株高产类胡萝卜素的海洋红酵母菌JMRY-1,通过形态学观察、生理生化测试及18S rDNA序列分析,将其鉴定为胶红酵母(Rhodotorula mucilaginosa)。进一步探究其生长特性及类胡萝卜素合成的最佳培养条件,结果表明:胶红酵母JMRY-1菌株最适生长温度为28 ℃;最适pH值为4.0;0~12 h为对数生长期,12~40 h为稳定期,40 h后进入衰亡期,28 h时生物量达到峰值(15.83 g/L)。通过正交试验优化类胡萝卜素合成条件,确定了最佳培养条件,即温度为28 ℃、时间为72 h、接种量为8%、转速为180 r/min,在此条件下类胡萝卜素产量达10.90 mg/L,较初始条件提升42.30%。
Abstract:
A marine red yeast strain JMRY-1 with high carotenoid production was isolated from natural seawater near Jimei District,Xiamen City,and identified as Rhodotorula mucilaginosa through morphological characteristics,physiological-biochemical tests,and 18S rDNA phylogenetic analysis. The biological characteristics and carotenoid synthesis conditions of the strain were systematically investigated. Results indicated that the optimal growth temperature and pH were 28 ℃ and 40,respectively,with a maximum biomass of 15.83 g/L at 28 h.Orthogonal experiments revealed that the optimal conditions for carotenoid production were 28 ℃, 72 h, 8% inoculation, and 180 r/min, yielding a carotenoid content of 10.90 mg/L, representing a 42.30% increase compared with initial conditions. 
