摘要:
从黄姑鱼转录组数据库中筛选出NK-lysin基因,命名为YdNkl-2。YdNkl-2基因cDNA全长600 bp,其中开放阅读框为456 bp,编码151个氨基酸。YdNkl-2具有一个信号肽和一个鞘脂激活蛋白B(saposin B)结构域。亚细胞定位结果显示,YdNkl-2分布于细胞质和细胞核。实时荧光定量PCR结果表明,YdNkl-2的mRNA广泛分布于黄姑鱼各组织,其中在鳃和脾脏中的表达量最高。经哈维氏弧菌感染后,头肾、肝脏和脾脏中的YdNkl-2表达量均明显升高,提示YdNkl-2在哈维氏弧菌感染黄姑鱼过程中发挥重要作用。
Abstract:
In this study,NK-lysin gene was cloned from the transcriptome database of Nibea albiflora,which was named YdNkl-2.The full length of YdNkl-2 cDNA was 600 bp,including an open reading frame(ORF) of 456 bp,encoding 151 amino acids.YdNkl-2 contains a signal peptide and a saposin B domain.The subcellular localization of YdNkl-2 was distributed in the cytoplasm and nucleus.The results of qRT-PCR showed that the mRNA transcripts of YdNkl-2 were ubiquitously distributed in all the detected tissues,and the expression was the highest in the gill and spleen.After infection with Vibrio harveyi,the expression of YdNkl-2 in head kidney,liver,and spleen increased significantly,suggesting that YdNkl-2 may play an important role in the process of V.harveyi infection.
