摘要:
UDP-葡萄糖焦磷酸化酶(UGPase)是红藻琼胶生物合成过程中的关键限速酶。以坛紫菜(Pyropia haitanensis)转录组测序获得的unigene序列为基础,采用RACE技术克隆获得了坛紫菜编码UGPase的基因序列:Phugp。序列分析结果表明:Phugp基因序列全长1734 bp,包含一个1530 bp的开放阅读框,所编码的多肽包含510个氨基酸,相对分子质量为56.190 ku,等电点为6.24,具有UGPase特有的底物结合位点和红藻UGPase保守的N端和C端多肽结合位点。基因表达水平的定量分析结果表明:在不同程度的高温和高光胁迫条件下,Phugp基因的表达水平均极显著下调,而在不同程度的失水胁迫条件下,Phugp基因的表达则呈现为逐渐上调的趋势,说明Phugp基因在坛紫菜遭受不同的胁迫条件时呈现为不同的表达模式,并可能在应答失水胁迫中发挥着应激调节作用。
Abstract:
UDPglucose pyrophosphorylase(UGPase),the key enzyme involved in agar biosynthesis of red algae.In this study,based on the unigene sequences obtained via whole transcriptome sequencing of Pyropia haitanensis,the fulllength of Phugp gene was obtained by rapid amplification of cDNA ends(RACE).The fulllength cDNA of Phugp gene comprised 1734 nucleotides and contained an open reading frame of 1530 bp,encoding a protein of 510 amino acid residues with the predicted molecular weight of 56.190 ku and theoretical isoelectric point of 6.24.The sequence of PhUGPase contains specific active sites of UGPase and conserved motifs of Nterminal and Cterminal in red algae UGPase.The expressions of the Phugp gene,as measured by realtime quantitative PCR,were significantly downregulated by hightemperature stress and high light stress,but upregulated by desiccation stress.These results suggested that the Phugp gene has different expression patterns in different stress and may play important roles in the response to desiccation stress.
